Pharmacology
Melatonin and melatonin agonists inhibit the release of dopamine from retina through activation of a site that is pharmacologically different from a serotonin receptor. These inhibitory effects are antagonized by the melatonin receptor antagonist luzindole, which suggests that melatonin activates a presynaptic melatonin receptor.
Dosage and Administration
How to use
Melatonin:
Take this medicine in the dose and duration as advised by your doctor. Swallow it as a whole. Do not chew, crush or break it.
Melatonin is to be taken empty stomach.
Adult Dose:
Oral

Adult: Usual dose 3-6 mg at bedtime.
Child Dose:
Oral
Disturbed biorhythms; Sleep disorders
Child: 2-3 mg at bedtime, increased after 1-2 wk to 4-6 mg if needed.
Max dose 10 mg.
Interaction
Quinolones; carbamazepine, rifampicin; fluvoxamine, 5- or 8-methoxypsoralen, cimetidine, cigarette smoking, oestrogens. Alcohol; zalepon, zolpidem, zopiclone; thioridazine, imipramine.
Contraindications
Pregnancy and lactation.
Side Effects
Common
- Headache
- Drowsiness
Increased seizure activity; drowsiness, headache. Disruption of normal circadian rhythm. May worsen symptoms for individuals with depression.
